Restriction digests of the clone give the following sizes (kb): HindIII--4.4, 2.7; BamHI--7.1; EcoRI--7.1; PstI--4.4, 1.65, 0.76, 0.3; AvaI--4.8, 2.0, <0.2. Most copies of the repeated sequence are in deletion interval 3B on Yp, but some are in interval 6 on Yq; genomic DNAs to be tested can be cut with anything, but Page et al. tend to use TaqI or EcoRI. |
